GUV counter

Re-counts and re-sizes giant vesicles from LUNA cell-counter frames. The instrument applies a contrast floor that misses the faint population, undercounting by 4.5–11×; this runs the same detection pipeline as the desktop GUV command — a matched filter against a bank of defocused-sphere templates — and reads the calibration straight out of the TIFF metadata, so concentrations stay on the instrument's own scale.

The defaults are the validated ones. Sensitivity is the only knob worth touching day to day.

Lower finds fainter, smaller vesicles and costs false positives: about 6 extra real objects per spurious one between 6.0 and 5.0.
matched recovers ~25 % more objects at the same false-positive rate.
The instrument's own gate was 3 µm.
30 or 60 µm depending on the protocol.

Frames are uploaded to the nanoscale.app server, analysed there and deleted automatically. Nothing is indexed, logged or kept beyond that window.
